The HPRT targeting methodology provides rigorous control over transgene genotype and inserts constructs into a desirable chromatin environment. Reproducible gene-appropriate expression levels are realized from most constructs.

This core facility will provide custom vectors to support convenient generation of HPRT targeting constructs, will receive client-generated constructs and derive ES clones bearing HPRT-docked transgenes. Constructs will be delivered into transgenic lines to support typical transgenic experiments as well as small to medium scale structure-function investigations.

  • consultation on construct design
  • Gateway shuttling of constructs into HPRT targeting vectors that are returned to client
  • transfection and recovery of multiple ES clones bearing each HPRT docked construct
  • preparation of frozen ES stocks
  • preparation of DNA from sub-clones that will be sent to the client for construct-specific genotype analysis or analyzed on site using client validated primers and PCR conditions

Cost: $2,500 per construct for CGDN members, $5000 per construct for non-CGDN members.
